I hope this makes sense.

Play a hold with custom monsters in it (good example holds to play around with include Growing and The 'Neatherhood from the RPG Holds boards). Start a New Game so that you have rooms visible on the map that are currently unvisited on this playthrough.

If you directly click on that room via the minimap, the custom monsters cannot have their stats and predicted damage viewed via the Battle screen and any _MyColor change is ignored. BUT, if you view the room by scrolling into it from an adjacent room using the arrow keys, then these things both work.

I believe there is code that essentially performs a "dummy run" of character scripting when you scroll into a room on "remote viewing" mode to correctly set the colour and stats of the monster. This code is not being run when you click on the room from the minimap, only when scrolling into the room.

Thanks for reporting this and for the clear description. Yes, this makes sense to me. You are correct that a "room load" operation is occurring when panning across rooms to initialize NPC stats, but it wasn't happening when clicking on a map room. Fixed in rev 327.
