I have cleared all room and now are running from the slayer to find the way to the next level. However, I am now in S4 with the slayer following me and there is no way out that I can see.

What is going on? Is this an error in the game or an error somewheres back when I finally met the slayer?

Is there any hope of getting out of 4S and/or 4S1E? See screen capture in the Attached File.

Okay so, I found the secret off this room while the Slayer was chasing me, and now he appears to, uh, not be here. And the door I need to go through is closed.

(This is all general thoughts without looking at the specific room.) The slayer's presence is tied to an entrance, so if you can get back to the way you would normally come in and re-enter, he should re-appear.

Otherwise, just restore to before you came into this room and went to the secret. Secret progress is tracked overall; you don't need to get all the secrets in a single play-through.

Are you playing the 5.0 update? If so, when you return to this room after the secret, a door to the east re-opens so that you can, as Tuttle said, go back and pick up the Slayer.

Pre-5.0, you would have had to go back round the level to the part where the Slayer first starts chasing you, or use Restore.
