You hit the orb (1) when the spawn timer is on 29.
The queen at (2) will move NE, the roach at (3) will move SW.
You can then head back to the checkpoint at (5) and send the queen out of the tunnel.
When you kill the queen at (4), the other queen will run north.
[EDIT: I had to fix the numbers up...was pointing at the wrong orb for (1).]
